NEUROPROTECTIVE EFFECTS OF KETAMINE, LAMOTRIGINE, AND KETOPROFEN IN RABBIT RETINAS AFTER PARS PLANA VITRECTOMY AND SILICONE OIL INJECTION

Introdução / Purpose

This study aimed to assess the protective impact of a combined treatment with ketamine (KTM), lamotrigine (LMT), and ketoprofen (KTP) on rabbit retinas in an experimental model of surgical aggression to the retina.

Material e Método / Methods

Thirty-six rabbits were divided into six treatment groups: saline, KTP, KTM+LMT, KTM+KTP, LMT+KTP, and KTM+LMT+KTP. All rabbits underwent pars plana vitrectomy (PPV) and silicone oil injection (SOI) in the right eye. The left eyes served as negative controls. Histological assessments were conducted to evaluate cell densities and structural changes in the ganglion cell layer (GCL), the inner nuclear layer (INL), and the outer nuclear layer (ONL). Apoptosis was evaluated using the TUNEL method.

Resultados / Results

The combination treatments demonstrated significant neuroprotective effects. The KTM+LMT combination showed protection in the GCL, while the KTM+KTP combination exhibited protection in the INL. The LMT+KTP combination provided protection in both the INL and GCL. Remarkably, the KTM+LMT+KTP combination demonstrated protection in the ONL and INL. Cell densities in the ONL, INL, and GCL were significantly higher in the treatment groups compared to the positive control group (p < 0.05).

Discussão e Conclusões / Conclusion

The combination of KTM, LMT, and KTP exhibited significant neuroprotective effects in the rabbit retina following PPV and SOI. These findings suggest the potential of this combination treatment as a therapeutic strategy for retinal pathologies. Further research is warranted to validate these results and further clarify the underlying synergistic mechanisms of action.
